Abstract
[Objective] The research aimed to establish loop-mediated isothermal amplification (LAMP) technology for rapid detection of Salmonella.[Method] The primers were designed according to the Salmonella-specific gene (invA) sequence as the target sequence.And the sequence was amplified under the isothermal conditions.The reaction conditions were optimized including Mg2+ concentration,reaction temperature and time etc.And Salmonella stains were detected and identified.[Result] The results indicated that the best amplification conditions was 62 ℃ for 40 min when Mg2+ concentration was 2.0 mmol/L.The sensitivity of LAMP detection of Salmonella stains was 7.8 cfu/ml.[Conclusion] The rapid method of LAMP detection of Salmonella was established.LAMP method was very rapid,sensitive and convenient,suitable for grass-roots generalization.This method built a technology platform for the detection of food-borne pathogens.
