Abstract
Objective To investigate effect of silencing survivin gene with siRNA on cell cycle,proliferation and apoptosis of SGC -7901 cell line,preliminary study on the mechanism.Methods Survivin -specific siRNA was designed and synthesized,small interfering RNA was transfected into SGC -7901 cells to knockdown the survivin expression.The cell growth inhibition was determined by MTT,mRNA levels of survivin and caspase - 3 was detected by RT - PCR,protein levels of survivin and caspase - 3 was detected by Western blot,protein levels of survivin was detected by immunohistochemical assay.The effect of survivin siRNA on cell cycle and apoptosis was assayed by flow cytometry(FCM).Results RNA interference efficiently suppressed the survivin expression in SGC -7901 cell line.After SGC -7901 cells transfected with survivin siRNA,proliferation rate was reduced(P0.05) and the expression of survivin protein levels decreased (P0.05);the expression of caspase - 3 was not significant in the mRNA levels(P0.05),but was increased in the protein levels.The percentage of apoptotic cells with survivin siRNA cells was higher.Conclusions Survivin siRNA can not only effectively decrease the expressions of survivin gene and protein,but also inhibit the cell proliferation in SGC -7901 cell.
