Determination of Amino Acids in Hirudo Extract by HPLC

A HPLC method was established for the determination of total amino acids and free amino acids in hirudo extract. An ODS column was used,with the mobile phase of 0.02 mol/L sodium acetate solution (pH 7.20) -0.1mol/L sodium acetate solution (pH 7.20)-methanol-acetonitrile by gradient elution. The detection wavelength was 338nm and 262nm. The recoveries of total amino acids and free amino acids were 97.8% and 98.3%,respectively. The RSDs were 1.76% and 1.90 %,respectively.
