Growth supressive effect of combined PTEN with antisense AKT2 on proliferation and apoptosis in C6 gliomas:an in vitro and in vivo study

Abstract

Objective To investigate the effect of combined PTEN with antisense AKT2 on proliferation and apoptosis of glioma cells. Methods Rat C6 glioma cells were transfected with PTEN-anti-AKT2 constructs, the expression and proliferative activitiy of PTEN-anti-AKT2 in cells were investigated. Parental C6 cells and C6 cells trasfected with PTEN-anti-AKT2 were implanted stereotactically into SD rats. The dynamic MRI and histopathological changes of the tumors and the expression of PTEN-anti-AKT2 were investigated. Results As compared with C6 cells, AKT2 expression was inhibited in transfected cell and PTEN on the contrary, proliferative activities were decreased in transfected cells. The survival time of rats in transfected group prolonged significantly than control group. Conclusions The combination of PTEN with antisense AKT2 gene transduction may be more suitable for clinical therapeutic trials of gene therapy for malignant gliomas.
