Efficient Purification of Lysozyme by One-step Cation Column Chromatography and Its Bacteriolytic Activity Assay

Abstract

OBJECTIVE:To study on how to purify lysozyme highly and efficiently from egg white by ion exchange chromatography.METHODS:The egg white was diluted and then precipitated by isoelectric precipitation to remove ovalbumin and ovomucin,etc.The supernant flowed into ethyloic-agarose gel for ion exchange chromatography.Samples of elution peak was collected and freeze-dried.SDS-PAGE and bacteriolytic turbidimetry were used to identify the purity and activity of the products,respectively.RESULTS:After isoelectric precipitation and one-step column chromatography,the purity of the products was over 95%,the specific activity was 17 600 U/mg,and the yield achieved 4.2 mg(lysozyme)per ml(egg white).CONCLUSION:One-step cation column chromatography can be applied to acquire high-quality lysozyme efficiently from egg white.
