Influence of Salvianolic Acid B on Expression of Bcl-2 and Bax Proteins in Rats with Focal Cerebra Ischemia Reperfusion

Abstract

Objective:To study the effects of sallvianolic acid B on the expression of bcl-2 and bax proteins in rats with focal cerebral ischemia reperfusion injury, and analyze the mechnism of action of sallvianolic acid B in preventing and treating ischemic cerebrovascular disease.Methods:24 healthy male Wistar rats were randomly divided into 3 groups with 8 rats in each group: sham-operation group, model group, sallvianolic acid B groups. The latter group infused corresponding drugs, once daily for 7 days. Then the intraluminal suture technique was used to establish the models of middle cerebral artery occlusion (MCAO) in rats except sham-operation group, for 2h of occlusion and 24h of reperfusion. At the end of the experiment, the immunohistochemistry and pathological image analytical technique were used to monitor the expression of bcl-2 and bax proteins after the focal cerebral ischemia reperfusion in brain tissue of the three groups, and ratios of bcl-2 and bax were compared. Results:In modle group, the expression of bcl-2 or bax proteins in brain tissue of rats was higher than those in sham-operated, the latter was increased more obviously; and ratio of both expressions was in imbalance. In sallvianolic acid B group, compared with model group, the expression of bcl-2 protein was increased, while the expression of bax protein was reduced; and ratio of both expression was recovered.Conclusion:Sallvianolic acid B could up-regulate the expression of bcl-2 protein, down-regulate the expression of bax protein simultaneously and recover the ratio of bcl-2/bax, which might be one of mechanisms of sallvianolic acid B preventing and treating ischemic cerebrovascular disease.
