Abstract

Objective To gain insight into the differentiation of the number,phenotypic profile and functionality of myeloid dendritic cells (DCs) in viral myocarditis.Methods Mouse bone marrow DCs were separated and cultured,then the expression of CD11c and CD40,CD80 on the DCs were detected through flow cytometry (FCM).Moreover,The levels of the protein of S100,CD40 and CD80 in the myocardium were detected by immunohistochemistry.Results Compared to normal group,DCs from VMC group were more susceptible to be mature in morphology.Meanwhile,FCM results showed that there was an increased number of DCs expressing CD11 c (P<0.01)and more fluorescence emitting from CD11c-CD40 or CD11c- CD80 double-positive cells(P<0.05)in the latter group.The cells among myocardium were to up-regulate of expressing S100 in VMC group than normal group,and CD40 or CD80 were positive too.Conclusions Our findings suggest that following more DCs generated from bone marrow,the number of them in heart is in- creased,which indicate a more mature DC phenotype after infected CVB3.
