Abstract
Objective: To investigate the protective effects of edaravone on lung in smoke inhalation injury in rats.Methods: Thirty-two male SD rats were randomly divided into four groups,Group A: normal control group(n=10),Group B: injured group(n=10),Group C:edaravone 8 h treated group(n=10),Group D: edaravone 24 h treated group(n=10).Thirty minutes after smoke inhaled injured,the rats in group C and group D were all injected intraperitoneally with edaravone(9 mg/kg),with the same volume the group B were injected intraperitoneally normal saline.The rats were treated by no means in group A.The blood samples were collected from abdominal aorta and the lung tissue of the rats in group C at 8 hours after injured,and the same to group D at 24 hours after injured.The levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6) in serum as well as the levels of malondialdehyde(MDA),nitric oxide(NO),inducible nitric oxide synthase(iNOS)and superoxide dismutase(SOD) in lung tissue were tested.Each right lung tissues were examined under an optical microscope.Results: Compared with group B,the TNF-α,IL-6,MDA,NO,iNOS levels decreased markedly in group C(P0.01),while the levels of SOD were increased(P0.05).The above changes were more evident in Group D(P0.01).Histopathological examination showed that inflammatory granulocytes and edema were improved in group C and group D compared with group B.Conclusion:Edaravone has protective effects on smoke inhalation injured,by inhibiting the production and release of free radicals and inflammatory mediators.
