Abstract

OBJECTIVE: To stude the apoptosis induced by thymope α1(Tsα1) in human hepatocelluar cancer SMMC-7721 cells. METHODS: Tsα1 inhibitatory SMMC-7721 cell’s growth by was studied MTT assay; morphological characteristics of apoptosis was observed by microscopy; cell apoptosis rate was measured by flow cytometry and agarrose gel electronphoresis; expression of bcl-2 was detected by flow cytometry. RESULTS: When the concentration of Tsα1 was 4 000 μg/mL, the inhibitatory rates of SMMC-7721 cells were (1.92±0.02)%, (3.27±0.03)% and (7.69±0.03)% at 24 h, 48 h and 72 h, respectively,P0.005. The apoptosis rates were (2.5±0.1)%, (18.3±0.2)% and (11.6±0.1)% at 24 h, 48 h and 72 h, respectively, P0.001. The inhibitatory rates of bcl-2 expression were (1.35±0.1)%,(2.2±0.01)% and (5.53±0.01)% at 24 h, 48 h and 72 h, respectively,P0.005. The morphological changes of apoptosis were fuound. CONCLUSION: Tsα1 can inhibit proliferation of SMMC-7721 cells, induce apoptosis in SMMC-7721 cells and inhibit expression of bcl-2.
