Genetic diversity in Festuca arundinaces based on random amplified polymorphic (RAPD) DNA analysis

Abstract

Fifteen varieties of F.arundinaces introduced from foreign countries were assessed for genetic diversity by RAPD DNA analysis using twelve primers selected from 60 arbitrary primers.They amplified a total of 85 DNA bands of which 59 DNA bands(69.41%) were polymorphic: the average number of polymorphic DNA bands amplified by each primer was 4.92.The range of Jaccard's genetic similarity coefficient among these varieties varied from 0.373 to 0.932 sa assessed by NTSYS-PC software.A DNA molecular dendrogram was established for 15 varieties of F.arundinaces based on UPGMA analysis of the genetic similarity matrix.Three groups were idnetified based on a standard of 0.68 similarity coefficient in the dendrogram,Triplet and Pixie each constituted a group and the other varieties clustered together as a third group.
