Abstract
Objective: To investigate the effects of edaravone on the acute renal ischemia/reperfusion(I/R) injury in dogs.Methods: Twenty-four adult male Mongrel dogs were randomly divided into three groups with 8 in each group: sham operation group,ischemia/reperfusion(I/R) group,edaravone treatment group.Blood samples were obtained for determination of blood creatinine(Cr) and urea nitrogen(BUN) concentrations before operation and 24,48,72 hours after operation.The dogs were killed on the third day after operation and left kidneys were removed for determination of SOD activity and MDA and MPO concentrations.The apoptosis in the nephridial tissue was assessed by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL),and apoptotic index(AI) was calculated.The changes of renal tissue were observed under microscope.Results: Blood Cr and BUN concentration in edaravone group decreased significantly than in I/R group in 3 days after operation.MDA and MPO concentrations were decreased,SOD activity was increased significantly and AI was decreased significantly in treatment group as compared with that respectively in I/R group on the 3rd day after operation(P0.05).Microscopic examination showed that the renal I/R injury was significantly attenuated by edaravone.Conclusion: Edaravone can attenuate renal damage induced by I/R through inhibiting oxidative stress,apoptosis and inflammation.
