Abstract
Objective Many studies have shown that abnormal modification of histones (deacetylation) closely correlated with the occurrence and development of glioma, the purpose of this study is to investigate the effect of trichomycin A (TSA) on the apoptosis of glioma cell line U251. Methods Different concentrations of TSA (0.2μmol/L, 0.4μmol/L and 0.8μmol/L) were used to treat glioma cell line U251, respectively. Western blot was used to detect the protein expression of caspase-3, acetylated hisitone H3 and H4 in U251 cells after TSA treatment. Cell apoptosis rate was detected by flow cytometry using Annexin V-FITC method. Results TSA treatment up-regulated the caspase-3 protein level significantly with a manner of dose-dependent ( P0.01). Flow cytometry showed that the apoptosis rate of U251 cells were up-regulated significantly after TSA treatment with a dose-dependent manner. Conclusion TSA treatment could up-regulate caspase-3 expression and promote the apoptosis rate of glioma cell line U251 with a dose-dependent manner.
