Abstract

[Objective] To evaluate the effect of aldosterone (ALD) on connective tissue growth factor (CTGF) mRNA expression in cultured rat mesangial cells. [Methods] ⑴ Mesangial cells were cultured in medium containing different concentrations of ALD (10-11、10-10、10-9、10-8、10-7 mol/L) for 72 h, while control cells were treated with vehicle only. The expressions of CTGF mRNA were detected by semi-quantitative RT-PCR. ⑵ Mesangial cells were stimulated with 10-7 mol/L ALD for 12, 24, 48, 72 h. The expressions of CTGF mRNA were detected by semi-quantitative RT-PCR. [Results] Semi-quantitative RT-PCR method showed that the expressions of CTGF mRNA were increased significantly in a dose-and time-dependent manner after stimulated by aldosterone in cultured rat mesangial cells.[Conclusion] ALD up-regulates the expression of CTGF mRNA in cultured rat mesangial cells. ALD may play a crucial role in the progression of chronic kidney disease.
