Abstract
GsP5CS,homologous with the GmP5CS gene encoding a Δ′-pyrroline-5-carboxylate synthetase was cloned from wild soybean(Glycine soja)by RT-PCR,with primers designed according to the sequence of GmP5CS.The largest open reading frame of GsP5CS gene has 2148bp in length and encoded a protein of 716 amino acid residues. The estimated molecular weight and isoelectric points of the putative protein was 77.9 kDa and 6.21,respectively.Hydropathy plot analysis indicated that the GsP5CS was hydrophilic.Blast result analysis showed that GsP5CS,GmP5CS,MsP5CS,MtP5CS,AtP5CS,BnP5CS,OsP5CS,TaP5CS shared a cluster. The phylogenetic tree shows that the relationship based on the homology is consistent with the morphological and taxonomic results.
