Association of human apo lipoprotein E gene polymorphism with serum high-density lipoproteins subpopulations

Abstract

Objective:To investigate human apolipoprotein(apo) E polymorphism and its relationship with serum lipids, apolipoprotein and serum HDL subpopulations. Method:apoE genotype was assayed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The subpopulations of serum HDL were determined by two-dimensional gel electrophoresis conjunction with immunodetection method. Result:apoE 3/3 genotypes and allele e3 frequency were both the highest with allele frequency e2(8.96%), e3(81.94%) and e4(9.10%) respectively. The genotype of apoE2 had higher serum TG,apoE levels and apoE/aopCⅢ ratio as compared with the genotype of apoE 3 or apoE 4 (P 0.05), but lower LDL-C and HDL 3b levels compared with the genotype of apoE 3 (P 0.05).In additional,we also found that in apoE 2 carriers HDL 2a have the tendency of increase compared with apoE 3 carriers,but no significant difference was observed. Conclusion:apoE gene polymoephism may related to the distribution of HDL subpopulations.
