Abstract
Three materials, polyvinyl alcohol(PVA),sodium alginate(SA)and PVA-SA,were used as support for entrapping carbamylation enzyme I in E.coli.The results showed PVA-SA immobilized particles had good mechanical strength and stability. For this groping PVA-SA composite vector, the optimum embedding conditions were determined as follows:PVA mass fraction of 8%,SA mass fraction of 1%,using 3% boric acid-1% calcium chloride with pH value of 6.7 as fixative. The recovery rate of enzyme was as high as 91.46% when the immobilized cells were prepared under above conditions.Having been conserved under 4℃ for a week and then for two weeks at room temperature, the immobilized cells still remained 90.76% enzyme activity of the original contrast, while free cells only remained 29.69%.
