Evaluation of different methods for glycosylated hemoglobin measurement

Abstract

Objective To evaluate three methods in glycosylated hemoglobin(GHb) measurements. Methods GHb was analyzed in linear range, recovery rate, precision, interference factor and normal reference range by ion-exchange high-performance liquid chromatography (HPLC) method (X), micro column affinity chromatography handwork method (Y 1), latex agglutination immunoassay (Y 2). Results The linear range of X 1,Y 1 and Y 2 methods were 2.8%-18%,3.6%-15.3% and 2.3%-15.5% respectively; The average recovery rate of three methods were all in 95%-104%; The average CV within and between-run were also less than 5%; No interference appeared when triacylglycerol concentration was up to 10.2 mmol/L, cholesterol up to 8.0 mmol/L, bilirubin up to 684 μmol/L and HbF up to 10.1%; The results of GHb measured by Y 1 method were significant higher than X method (P0.01), the regression equation Y 1=1.15X-0.17(r=0.86,P0.01). There was also a marked difference in GHb results between X and Y 2 method (P0.05). The results of GHb measured by Y 2 method were lower, Y 2=1.04X-0.69(r=0.92,P0.01). The normal reference value of X,Y 1,Y 2 methods were 4.3%-6.7%, 3.5%-8.3%, 3.6%-6.7%. Conclusions Comparing with HPLC method, the result of micro column affinity chromatography handwork method is higher and the result of latex agglutination immunoassay is lower.
