Abstract

Objective:Determine the efficacy of Tong Xin Luo(TXL)on the apoptosis and the activity of Caspase-3 in the endothelial cell under hypoxia.Method:the Bovine aortic endothelial cells(BAEC) were divided into three groups and incubated with normal medium,hypoxia medium,hypoxia medium+TXL(50ug/ml)for 24h respectively.Apoptosis were measured with TUNEL.Western blot and the kit of Caspase-3 activity determination were used to measure the expression and the activity of Caspase-3 in BAEC.Result:TXL can decrease the percent of apoptotic cells in BAEC induced by hypoxia with mark significance (11.84±1.4% vs 28.85±2.46%, P 0.05).Although TXL had no effect on the expression of Caspase-3 after hypoxia stimulation,TXL could abolished the increase of the activity of Caspase-3 in BAEC induced by hypoxia(41.2±5.5 VS 67.8±7.2 U/ml, P 0.05).Conclusion:The inhibition of the activity of Caspase-3 might be the main mechanism of TXL decrease endothelial cell apoptosis induced by hypoxia.
