Abstract
Objective To investigate the effects of curcumin on enhancing the sensitivity of leukemia U937 cells to TRAIL and to explore the possible molecular mechanisms. Methods MTT assay was used to measure the influence on cytotoxic activity of curcumin or( and) TRAIL against leukemia cell line U937 in vitro, apoptosis detected by flow cytometry. Results Curcumin has shown obviously cytotoxic effects on leukemia U937 cell and in dose-dependent manner while TRAIL has no cytotoxic effects. Different concentrations of curcumin would significantly up-regulate the expression of DR5. Conclusions Curcumin could enhance the sensitivity of leukemia U937 cells to TRAIL and its apoptosis mechanism may be correlated with the up-regulation of DR5.
