Abstract
Short tandem repeats(STR ) or microsatellite are the repetitive sequences with a repeat unit of 2-6 hp dispersed in the whole human genome. The length of STR are quite unstable and highly polymorphic with regard to the number oftheir repeat unite. However, they segregate in Mendel's fashion. That unique advantage and it's potential characteristic for automatic analysis have made STR the second-generation DNA marker. In this report we initially deve4op a method for isolating STR from the Short arm of Chromosome 5.Triplet degenerate probes were designed to suit the purpose of detecting all the trinucleotide repeats in human genome. One positive cosmid was obtained and subcloned into pBluscript by hybridizations. At last, we get a compound STR after sequencing.
