Experimental study of apoptosis effects of diallyl trisulfide on human leukemia cells HL-60

Abstract

Objective To study the effects of diallyl trisulfide(DATS) on apoptosis of human leukemia cells HL-60 and the probable mechanism involved.Methods The HL-60 cells were co-incubated with DATS at 30 mg/L for 48 hours.Morphological changes of HL-60 cells were observed by light microscope.The apoptosis index of cells was measured by flow cytometry(FCM).Immunohistochemical staining was used to detect the expressions of Bcl-2 and survivin in HL-60 cells.Results DATS could make cells HL-60 morphologic features of apoptosis.The apoptosis indexes of cells HL-60 with DATS at 0,30 mg/L for 48 hours were significantly different(P0.05).Compared with control group,the expressions of Bcl-2 and survivin in the test groups decreased distinctly.Conclusions DATS may induce apoptosis of HL-60 cells.The mechanism is probably correlated with down regulation of Bcl-2 and survivin in HL-60 cells by DATS.
