Detection of intracellular compatible solute ectoine in halophilic bacteria from Qinghai Lake by high performance liquid chromatography

Abstract

In this study,we isolated 35 bacterial strains in 20 water samples from Qinghai Lake.The results from salt resistant gradient experiments showed that moderate activity halophiles accounted for about 65.7%,while weak activity halophiles were about 25.7%.Nonhalophile and halotolerant were relatively low,only about 8.5%.The accumulation of ectoine in moderate activity halophilic bacteria was analyzed by TLC and HPLC,and the results revealed that compatible solute ectoine accumulated in most moderate halophiles.Under the optimum growth salinity,the amount of ectoine in QHL1,QHL5,QHL10,QHL26 and QHL28 reached 258.60,325.47,208.8,292.31,295.13 μg·mL-1,respectively.Finally,five wild strains with high ectoine production capability were obtained.
