Rapid PCR Detection of Salmonella in fecal samples of animal

Abstract

In order to establish a rapid,sensitive and accurate detection technique for Salmonella in animal feces,set of primers were designed according to stn gene in Salmonella to detect Salmonella of various serovars by PCR amplification.The method's sensitivity,specificity and the detection limit in the artificial feces samples were studied.conventional culture technique was used to demonstrate the validity of this method.The rusults showed that primers used in this experiment was specific to Salmonella and the band of 260 bp was obtained.The 43.85 pg DNA of Salmonella was detected by PCR.The detection sensitivity was 1 cfu/mL in pure culture.The detection limit of artificially contaminated feces samples was 103 cfu/mlL and didn't need to enrichment.While the detection limit was 1 cfu/mL using enrichment procedure.The PCR method detected Salmonella in 18 out of the 250 samples analyzed,and conventional culture technique support the result.The PCR method was suitable for the detection of Salmonella in feces.
