Abstract
Objective To study the inhibitory effect of RNA interference (RNAi) on survivin expression in bladder cancer cell line, 5637. Methods The 5637 cclls were transfected with synthetic survivin sequence-specitic siRNA by lipofectamine. The expression of survivin mRNA was detected by real-time quantitive PCR. Apoptosis of the transfected cells was examined by flow cytometry. Results The 5637 cells was transfected with dsRNA-survivin,and the expression of survivin mRNA was suppressed markedly. Compared with the control,the inhibitory rate of survivin mRNA was 29% and 53% at 24 and 48 hours after transfection with the 40nmol/L specific siRNA,when the density of siRNA was 100nmol/L, the inhibitory rate of survivin mRNA was 46%and 86%,respectively.Apoptosis was identified in the transfected 5637 cells.Conclusion The expression of survivin at transcriptional levels in the 5637 cells transfected with siRNA is markedly inhibited with an induction of apoptosis.
