Effect of interference RNA on survivin expression in 5637 bladder cancer cells

Abstract

Objective To study the inhibitory effect of RNA interference (RNAi) on survivin expression in bladder cancer cell line, 5637. Methods The 5637 cclls were transfected with synthetic survivin sequence-specitic siRNA by lipofectamine. The expression of survivin mRNA was detected by real-time quantitive PCR. Apoptosis of the transfected cells was examined by flow cytometry. Results The 5637 cells was transfected with dsRNA-survivin,and the expression of survivin mRNA was suppressed markedly. Compared with the control,the inhibitory rate of survivin mRNA was 29% and 53% at 24 and 48 hours after transfection with the 40nmol/L specific siRNA,when the density of siRNA was 100nmol/L, the inhibitory rate of survivin mRNA was 46%and 86%,respectively.Apoptosis was identified in the transfected 5637 cells.Conclusion The expression of survivin at transcriptional levels in the 5637 cells transfected with siRNA is markedly inhibited with an induction of apoptosis.
