Abstract
Objective To study the effect of probucol on oxidation and the activity of proliferating cell nuclear antigen(PCAN) and NFkBp65 in atherosclerosis. Methods 24 male New Zealand white rabbits were randomly devided into 3 groups: normal group, high fat diet group and probucol group. After 10 weeks, the level of serum triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL),superoxide dismutase(SOD) and malondialdehyde(MDA) were measuered. The expression of the PCAN and NFkBp65 in aorta were measured by the method of immunohistochemistry. Results Compared with those of the normal group the level of serum TG,TC and LDL-C dramatically increased(P 0.01),but the level of serum HDL-C decreased(P 0.01)in the high fat diet group.Compared with that of the high fat diet group the level of serum TG in probucol group decreased, but had not reached statistical diference(P 0.05).In the probucol group, the level of serum TC and LDL-C decreased(P 0.01) and the level of serum HDL-C also decreased(P 0.05)compared with those of the high fat diet group. The level of serum SOD was lower and the level of serum MDA was higher in high fat diet group than those of the normal group(P 0.01),while the level of serum SOD was higher and the level of serum MDA was lower in the probucol group than those of the high fat diet group(P 0.01).In the high fat diet group the exepression of PCNA and NFkBp65 were advanced compared with those of the normal group(P 0.01),but they were reduced in the probucol group compared with those of the high fat diet(P 0.01).The expression of PCNA was positively correlated with the expression of the NFkBp65 in the plaque of the aorta(r =0.916,P 0.01).Conclussion Probucol can relieve the oxiditive injury and repress the activity of NFkB so as to inhibit the proliferation of smooth muscle cells.
