Abstract
Objective To explore the impact of smoking on Fibulins family and the intervention role of Astragaloside in fibroblast.Methods The morphological changes of fibroblasts after 24 hours' treatment with cigarette smoke extract(CSE) were observed with inverted microscope.The inhibitory effect of CSE at different concentrations and the change after intervention with Astragaloside on fibroblast were determined by the MTT test.The mRNA expressions of Fibulin-3,-4,-5 were detected by fluorescence quantitative PCR assay.Results Light microscopy showed that fibroblasts lost their normal morphology after exposed to CSE.MTT assay showed a significant inhibition of fibroblasts cultured with CSE in vitro and the inhibition is in a dose-dependent manner.The proliferation rate of fibroblasts rises in a dose-dependent manner when astragaloside was added to the culture after cigarette exposure,this promotive effect is most obvious at 100 μg/mL.RT-PCR showed that Fibulin-3,-4,-5 expression levels were significantly decreased after the treatment with CSE and their expressions were upregulated after adding astragaloside at 100 mg/L concentration(P0.05).Conclusion Smoking could cause abnormal elastic fiber assembly through affect the expression of Fibulins family resulting in elastosis of the skin and Astragaloside has the protective effect on it.
