Development of HLA B2704 Transgenic Mice

Abstract

Objective To develop HLA B2704 transgenic mice.Method HLA B2704 gene was introduced into mice by microinjection.The presence of HLA B2704 was confirmed by PCR in the founder mice and their progeny.Further identification and quantitation of HLA B2704 gene were determined by Dot blot hybridization and Southern blot hybridization.Immunohistochemistry staining and flow cytometry were carried out to detect the expression of HLA B2704 antigens.Results Ten founders contained HLA B2704 gene.Seven of 37 (18 9%) mice from C57BL/6×Kunming and 3 of 64 (4 6%) from Kunming×Kunming background integrated HLA B2704 gene.Statistical analysis showed that there was significant difference between the two groups ( P 0 05).HLA B2704 antigen was expressed within the cytoplasma of skin and gastrointestinal tract of examined mice.Conclusion HLA B2704 transgenic mice have been obtained.C57BL/6×Kunming F 1 is better candidate for the development of transgenic mice.
