Abstract
Objective:To study the expression of TLR2 and TLR4in HSP children and its correlation with the Th1 and Th2type immune response.Methods:15cases of HSP children without renal failure were enrolled as NHSPN group,15 cases of HSP children with renal failure were enrolled as HSPN group,and 15 cases of healthy children were enrolled as healthy control group.Peripheral blood mononuclear cells were isolated,PCR was used to detect mRNA expression of TLR2 and TLR4,and flow cytometry was used to detect protein expression of TLR2 and TLR4;serum was collected and ELISA was used to detect IFN-γand IL-4contents.Results:(1)Serum IFN-γcontents and IFN-γ/IL-4ratios of HSPN group and NHSPN group were lower than those of healthy control group,and IL-4contents were higher than those of healthy control group;serum IFN-γcontents and IFN-γ/IL-4ratios of HSPN group were lower than those of NHSPN group,and IL-4contents were higher than those of NHSPN group;(2)mRNA contents:positive cell rates,fluorescence intensity of TLR2 and TLR4in peripheral blood of HSPN group and NHSPN group were higher than those of healthy control group;TLR2and TLR4 expression in peripheral blood of HSPN group was higher than that of NHSPN group;(3)fluorescence intensity of TLR2 and TLR4in peripheral blood mononuclear cells was negatively correlated with serum IFN-γcontents and positively correlated with serum IL-4contents.Conclusion:TLR2and TLR4 expression increases,and Th1/Th2 function is in disorder in peripheral blood of HSP children;overactive TLR2 and TLR4may be involved in weakened function of Th1 and enhanced function of Th2.
