Abstract

Objective:To investigate the effect of valsartan on ventricular remodeling and angiotensin Ⅱ and aldosterone in non-infarcted myocardium of left ventricle after myocardial infarction (MI). Methods:The rat model of MI was established by ligating the left coronary artery in female Wistar rats. Three hours after the ligation,21 survived rats were randomly divided to MI group,valsartan group,and sham operation group. The rats were lavaged with 15 mg·kg-1·d-1 valsartan in valsartan group and tap water in MI and sham operation groups for 4 weeks. The left ventricular transverse diameter (LVTD),which indicates the changes of left ventricular structure,was detected. The collagen volume fraction (CVF) in non-infarcted myocardium was detected after HE staining,and the contents of angiotensin Ⅱ and aldosterone in non-infarcted myocardium were determined with radioimmunoassay. Results:Compared with sham operation group,LVTD,CVF,and the contents of angiotensin Ⅱ and aldosterone were all significantly increased in MI group. LVTD,CVF,and the contents of angiotensin Ⅱ and aldosterone in valsartan group were all significantly lower than those in MI group. Conclusion:Valsartan may improve the ventricular remodeling after MI by inhibiting the overexpression of angiotensin Ⅱ and aldosterone.
