The discussion of the VEGF mechanism in ascites formation and peritoneal metastasis of ovarian cancer

Abstract

Objective By detecting expression of VEGF and its receptor Flt-1 protein and peritoneum of angiogenesis in ovarian cancer tissues,to discuss the mechanism of VEGF in ascites formation and peritoneal metastasis pots of ovarian cancer.Methods Detect VEGF,Flt-1 protein expression and their situation and angiogenesis in omentum and peritoneum of 43 cases ovarian cancer patients hospitalitied in the second medical hospital of Jilin University by SP immunohistochemistry.Results 1.In ovarian cancer,the patients,who was in stage Ⅲ-Ⅳ,whose ascites was above 500 ml or those had pots of peritoneal metastasis got higher rate of positive expression of VEGF and its receptor Flt-1 than stage Ⅰ-Ⅱ,whose ascites was below 500 ml,or those without pots peritoneal metastasis(P0.05);The positive rate of VEGF and Flt-1 expression in ovarian cancer Patients' ovarian tissue,was higher than benign tumors and the normal control group(P0.05).In ovarian cancer patients's ovarian tissue,cancer cells,vascular endothelial cells and lymphocytes could express VEGF,but in benign ovarian tumors and normal ovarian tissue of ovarian control group,the main VEGF-expression cells were vascular endothelial cells.Flt-1 were mainly expressed by vascular endothelial cells in all kinds of ovarian tissue,and some ovarian cancer cells also express Flt-1.The microvascular density(MVD) of ovarian tissue and VEGF expression in lymphocytes in ovarian cancer patients who with peritoneal metastasis were higher than those without(P0.05);Both in omentum and peritoneal tissues,the positive expression rate of VEGF in those existed cancer metastasis were higher than those without cancer metastases(P0.05).Conclusion 1.In ovarian cancer patients's ovarian tissue,cancer cells,vascular endothelial cells and lymphocytes all can express VEGF;2.Ovarian cancer cells not only secreted VEGF themselves,but also can influent the cells around the tumor,and then achieved the local ovarian cancer invasion、metastasis and proliferation.3.VEGF was secreted into blood serum and ascites,then promoted cancer angiogenesis,and formed a massive ascites,and resulted in abdominal and distant metastasis.
