Expression and Significance of P53 and VEGF in Serum and Xenograft of SKOV3 Tumor-Bearing Nude Mouse Model of Adverse Psychological Stress

Abstract

Objective:To investigate the effects of adverse psychological stress on P53 and vascular endothelial growth factor(VEGF)protein levels in serum and xenograft of SKOV3 tumor-bearing nude mice.Methods:The adverse psychological stress models of nude mice bearing ovarian cancer were established.The serum levels of P53 and VEGF were detected by ELISA in mice of normal control(A),simple stress(B),simple tumor(C) and tumor-bearing stress(D) groups(n=10 for each group).Western blot was performed to detect the expression of P53 and VEGF protein of xenograft in C and D groups.The P53 and VEGF protein expressions were also detected by immunohistochemical examination in 32 xenograft samples of D group and 24 samples of C group.Results:The serum levels of P53 and VEGF were not significantly different between B and A groups(P 0.05).The serum levels of P53 and VEGF were higher in C group than those of A group(P 0.01).The serum levels of P53 and VEGF were higher in D group compared with those of B and C groups(P 0.05 or P 0.01).Western blot showed that the average density ratios of P53 and VEGF were significantly higher in D group than those in C group(P 0.05).The results of immunohistochemistry showed that positives expression rates of P53 protein were 87.5% and 62.5% in D group(n = 32) and C group(n = 24),while VEGF protein were 93.8% and 70.8%,the differences were significant(P 0.05).Conclusion:The adverse psychological stress may accelerate the progress of SKOV3 tumor,which may be related to the expression of P53 and VEGF.The expressions of P53 and VEGF are expected to become the molecular targets for psycho-oncology study.
