Abstract

Objective To discuss the LC/MS/MS method for determination isoscutellarin concentration in human plasma by using scutellarin. Methods Using Agilent SB-C18 column(4.6 mm×150 mm,5 μm),the mobile phase consisted of methanol-water-formic acid(55:43:2).Using electrospray ionization source(ESI) and Selected reaction monitoring(SRM) mode,LC/MS/MS was applied to werify scutellarin,isoscutellarin and its aglycones, which were operated in negative ion mode and product ion mode.Ions fragment reaction were m/z 463a287 and m/z 287a123 respectively.Results The structure of scutellarin and its metabolized produces were deduced based on the chromatography and MS data,and isoscutellarin concentration in human plasma was determined.Conclusion The LC/MS/MS Method for Determination Isoscutellarin Concentration in Human Plasma by using Scutellarin can be applied.
