Extraction Process of Polysaccharide from Lonicera japonica with Compound Enzyme Treatment and Its Antioxidant Activity

Abstract

In order to optimize the compound enzymatic extraction technology of polysaccharide from Lonicera japonica and and investigate its antioxidant activity. The experiments used polysaccharides extraction rate as response value, and used enzymatic hydrolysi(cellulose, pectinase, papain), the ratio of water to material, pH, extraction temperature and extraction time as experimental factors to optimize the polysaccharide extraction conditions from Lonicera japonica. Antioxidant activity of polysaccharides was measured by DPPH and O2-· free radical elimination method. The best ratio of compound enzyme was 2.0% cellulase, 2.0% pectinase, 0.5% papain. The optimum conditions were as follows, the ratio of water to feedstock was 25 m L/g, pH4.5, extraction temperature was 50 ℃, extraction time was 60 min. Under the optimal conditions,the experimental extraction rate was 12.36%. IC50 of DPPH and O2-· were 0.811 mg/m L and 1.363 mg/m L, respectively.Antioxidant activity of sample polysaccharides was weaker than those of vitamin C. The optimum compound enzymatic extraction technology of the polysaccharides from Lonicera japonica was convenient and feasible, the polysaccharides extraction rate was higher than other extraction methods, and the extracted polysaccharides had good antioxidant activity.
