Abstract
Some related factors influencing explants callus rate in the process of cotton genetic transformation mediated by Agrobacterium tumefaciens were studied.The results showed that the optimal concentration of Kanamycin(Kan) was 50 mg/L or 75 mg/L,the ability of disseminating of Agrobacterium tumefaciens EHA105 was more powerful,the optimal material of explant was hypocotyl of 7-days-old seedling,the pre-incubated hypocotyl explants before disseminated by Agrobacterium tumefaciens were more favorable to the formation of embryogenic callus,and the optimal period of disseminating was 3 minutes.
