A Flaking Method of Observing the Rolling and Stereo Shapes of Pollen under Optical Microscope

Abstract

The study aimed to introduce a flaking method of using the immersion oil to observe the rolling and stereo shape of pollen,as well as the method of reorganizing common microscope(bright field microscope) into dark field microscope and the observation result.The microscope observation should be conducted at once after the flaking was finished by spraying botanic pollen on the microscope glass slide,covering the cover glass and knocking it for 1~2 times with soft things such as eraser on the pencil,then dropping a small drop of immersion oil on the outside or corner of cover slip,with the oil diffusing along the gap between microscope glass slide and cover glass into inside.The rolling course,stereo shape,hierarchy structure and surface ornamentation of pollen were easier to observe and record visually under 40-fold microscope by flaking pollen with immersion oil.A multi-use microscope could be made by doing some structural remodeling on the microscope.With continuous upgrade of optical microscope,it would have more and more advantages which couldn't be replaced by electrical microscope on aspects such as observing pollen shape and structure information.
