Abstract
Objective To establish two-dimensional gel electrophoresis with high resolution and reproducibility for proteomic study of pancreatic cancer.Methods Normal pancreas and surgically resected pancreatic cancer specimens and pancreatic cancer cell lines were homogenized with lysis solution. Proteome was detected by two-dimensional gel electrophoresis.Results Protein compositions of tissues and cells were well abstracted by increased protease inhibitors and pharmalytes. Two-dimensional gel electrophoretograms were clear and reproductive. Conclusions Sample preparation and electrophoresis condition were the key events for two-dimensional gel electrophoresis.
