Effect of High-D-Glucose on Cell Cycle and Apoptosis in Cultured Vascular Endothelial Cells

Abstract

Objective:To investigate the effects of high glucose on cell cycle and apoptosis in the cultured human umbilical vein endothelial cells (HUVECS). Methods: HUVECS were treated with different concentrations of glucose (5,20,40 mmol/L) for 24,48,72 hours.The cell proliferation curve was made to observed the inhibition effects of glucose on HUVECS growth.Cell cycle and apoptosis were measured by flow cytometry. Results: High-glucose(20,40 mmol/L) significiently inhibited HUVECS proliferation(P0.01) and accelerated cell apoptosis in a dose and time dependent manner than the low-glucose(5mmol/L) group(P0.01).In the meanwhile ,the percent of the cultured cells in G 0/G 1 phase in high glucose were increased, the percent of S and G 2/M phase were decreased (P0.05) .Conclusion: High-glucose(20mmol/L?40mmol/L) may inhibit the cell proliferation and accelerated cell apoptosis of HUVECS. Endothelial cells were possibly arrested in G 1/S checkpoint.
