La-tPA expression determination by RT-PCR in mammary gland of transgenic mice

Abstract
The vector of expression La tPA in mammary gland of transgenic animal was constructed by using sheep β lactaglubin(BLG) gene 5′ control region.6 transgenic mice were produced by microinjection method and PCR and Southern blot identification.In order to investigate precisely expression regulation of transgene,foreign gene transcript was determined by RT PCR during secrete milk period 1 20 days in transgenic mice with whey acid protein (WAP)gene as internal control.The results showed that human La tPA mRNA level reached the highest in 10 15 days and the lowest in starting and ending of secrete milk in mammary glands.
