Abstract
Objective To explore the therapeutic effect of matrine on liver fibrosis and its influence on the expression of IL-10,TGF-β1 and type Ⅰ collagen in hopes of providing experiment data for the prevention and cure of liver fibrosis.Methods 35 SD rats were divided into normal control group,carbon tetrachloride(CCl4)-induced liver fibrosis model group and matrine intervention group randomly.Immunohistochemistry,HE and Masson staining were used to measure the expression of type Ⅰ collagen in liver tissue.The level of IL-10 and TGF-β1 in serum was measured by enzyme linked immunosorbent assay(ELLISA).Results The average OD values of type Ⅰ collagen were 0.298,0.313 and 0.503 in the normal control group,matrine group and liver fibrosis group,respectively.The level of IL-10 were 1 412.38 pg/L,1 311.81 pg/L and 877.03 pg/L,the level of TGF-β1 were 491.29 pg/L,665.36 pg/L and 959.09 pg/L,respectively.The difference had statistical significance between normal control group and liver fibrosis group,and between matrine group and liver fibrosis group(P0.01).Conclusion In this CCl4-induced rat model,liver fibrosis was relieved by the administration of matrine.TGF-β1 and IL-10 may be use to evaluate the clinical intervention in the treatment of liver fibrosis.
