Abstract

Objective To observe the effect of ketamine on expression of GAP-43 in 7-day-old infant SD rat hippocampal neural cell.Methods The 7-day-old SD rats were injected with ketamine either 25mg/kg(group K_1)or 50mg/kg(group K_2),the rats were killed 24h after injected.The GAP-43 protein of SD infant rat's hippocampal was tasted by using immunhistochemistry stainning and Western blotting.Results 24h after treatment with ketamine,the number of neural cell that expressed the GAP-43 protein by immunhistochemistry stainning were decreased,group k_0 was 85.4±15.2/mm~2 ,group k_1 was 67.6±11.5 / mm~2,group k_2 was 36.6±9.7 / mm~2.The grey values of GAP-43 protein by Western blot were decreased too,group k_0 was 186.375±7.45,group k_1 was 165.754±7.012,group k_2 was 158.852±8.721.Conclusion 24h after treatment with ketamine,the expression of GAP-43 protein is cutted down in hippocampal neural cell,maybe it is involved in neurotoxicity treatment with ketamine.
