Abstract

Objective To evaluate expressions of NF-кB and ERK-1 mRNA in rats with liver fibrosis and the influence of curcumin to them.Methods Liver fibrosis rats were induced by carbon tetrachloride (CCl4). Rats were divided into three groups:normal group,model group and curcumin group. The expression of alpha-Smooth Muscle Action was detected by immunohistochemistry. The expressions of nuclear factor kappaB (NF-кB) and extracellular signal-regulated kinase (ERK-1) mRNA were determined by RT-PCR.Results The expression of alpha-Smooth Muscle Action was significantly lower in the rats treated with curcumin than that in the model group(P0.05). The expressions of NF-кB and ERK-1 mRNA were significantly higher in model group,but obviously lower in curcumin group(P0.05).Conclusion Curcumin is able to inhibit the activation of hepatic stellate cell through suppressing the expression of alpha-Smooth Muscle Action and expressions of NF-κB mRNA,and it also can inhibit the proliferation of hepatic stellate cell through suppressing the expression of ERK-1 mRNA.
