Abstract
Objective To investigate the effects of 6-hydroxydopamine (6-OHDA) on free radical, dioanube (DA),GDNF and apoptosis in rats. Methods Using biochemical, immunohistochemical and TUNEL techniques, the changes of apoptosis, free radicals, dopamine content and GDNF expression in substantia nigra and striatum were observed after stereoscopically injecting 6-OHDA to right side of the mesencephic ventral tegmental area(VTA) and substantia nigra pars compacta(SNpc). Results The free radical and number of apoptosis in substantia nigral were increased but the GDNF expression and dopamine content in striatum were decreased compared to control group (P0.05).Conclusion Free radical,GDNF and apoptosis contribute to the pathogenesis of 6-OHDA-induced PD in rat.
