THE TREATMENT EFFECT OF EARLY-USED HIGH-DOSE VITAMIN C ON RATS WITH ACUTE HEMORRHAGIC NECROTIZING PANCREATITIS(AHNP)

Objective To observe the effect of early used high dose Vitamin C for the treatment of acute hemorrhagic necrotizing pancreatitis (AHNP) rats and to study its mechanism.Methods Seventy two rats with AHNP were divided into 3 groups,24 rats into each.And 8 healthy rats were taken as normal group.Group A:NS (5 ml/kg) was administrat ed through vein.Group B:NS(5 ml/kg) and Vitamin C (15 mg/kg) were administrated through vein.Group C:NS (5 ml/kg) and Vitamin C (150 mg/kg) were administrated through vein.8 and 24 hours after the induction of rats,8 rats of each group were sacrificed and blood samples were taken for the measurement of serum amylase,lipase,SOD,PV C,endo toxin,MDA,TNF αand IL 6,and pathological study of pancreas was performed.The living rate in 3 days of other 8 rats of each group was observed.Results The living rate of each group was normal group (100%,8/8),Group A (0%,0/8), Group B (12.5%,1/8),Group C (50%,4/8).The 4 aspects of pathological scores of pancreas were much higher in AH NP rats than in the normal group.The 4 aspects of pathological scores of pancreas were lower in Group C than in Group A and Group B.The serum level of amylase,lipase,endotoxin,MDA,TNF αand IL 6 were much higher in AHNP rats than in the normal group and lower in Group C than in Group A and Group B.The serum level of SOD and PV C were lower in AHNP rats than in the normal group,while certainly higher in Group C than in Group A and Group B.Conclusion It is quite effective to use high dose Vitamin C for the treatment of AHNP rat in the early time,which is able to increase the level of SOD and PV C,inhibit synthesizing and excreting panreatic enzyme, keep out free radicals and cytokines in the body,improve the micro circulation of pancreas,reduce the pathological scores of pancreatic hemorrhage and necrosis and the ruin of ultramicrocirculation of pancreas.
