Induction of apoptosis of human Burkkit lymphoma cell lines Raji by X-ray irradiation

Abstract

Objective To investigate the effects of X-ray irradiation on the proliferation and apoptosis of human Burkkit lymphoma cell line Raji.Methods The cells were irradiated with various doses of irradiation.After irradiation,the morphology of the irradiated cells was observed under microscope.Cell cycle and apoptosis were assessed by flow cytometry.Results At 48 hours after irradiation(16 Gy),the percentage of growth inhibition was(92.67±1.20)%.The proportion of early apoptotic cells induced by 8 Gy irradiation at 48 h was greatest(42.04±3.62)%.At 24 hours after 8 Gy irradiation,the proportion of cells(57.86±3.31)% arrested at G2/M-phase was significantly greater than the control group(14.54±2.32)%.Results from the clonogenicity assay showed that the number of colony after 8 Gy irradiation(5.33±2.40) was lower than the control group at day-7(116.67±20.28) and day-14(263.33±20.27).Conclusion X-ray irradiation can inhibit the proliferation,induce cell cycle arrest,and induce apoptosis of Raji cells.X-ray irradiation may also inhibit the growth of cancer stem cells.
