Abstract
Objective To observe the effects of resveratrol on expression of NF-κB and VEGF of human hepatocellular carcinoma cell line HepG2 in vitro in order to explore the possibility of its inhibition on proliferation initially.Methods The frozen HepG2 was cultured for generation and treated with resveratrol of different concentration for 24h respectively.RT-PCR was used to detect the expression of VEGF mRNA,and the NF-κB activation and protein expression levels were evaluated by electrophoretic mobility shift assay(EMSA).Results Resveratrol inhibited both VEGF mRNA expression of HepG2 cells with obvious dose and time-dependent effects(P0.05).Moreover,NF-κB activity of 20ng/ml TNF-α-induced HepG2 were decreased by resveratrol with a dose dependent manner(P0.05).Conclusion Resveratrol could inhibit expression of VEGF of HepG2 and make NF-κB activity down-regulating.So it is presumed that resveratrol may suppress proliferation of hepatocellular carcinoma.
