Abstract
By using bovine serum album in (BSA) as an intermediary, conjugate of methotrexate (MTX) with monoclonal antibody MGb2 (IgG) , against gastric cancer was prepared with improved tumour specificity. The linking of MTX with BSA was performed, without protect ion of the thiol group of BSA, by using MTX active ester prepared by treating MTX with DCC and NHS. The MTX-BSA preparation was coupled to MGb2 with iodoacetyl group introduced in advance. The molar ratio of IgG: BSA: MTX was l: 1. 5:30 in the conjugate. In vitro studies indicated that the conjugate showed higher selective cytotoxic effect on gastric cancer cell line than that on controlled fetal lung cell line.
