The association study of MTHFR gene polymorphism with depression

Abstract

Objective To investigate the relationship between C677T polymorphism of MTHFR gene with depression.Methods A total of 94 patients with depressive disorder in study group and 98 normal health controls in control group were involved in this study.The serum levels of homocysteine were determined by fluorescence polarization immunoassay(FPIA).Polymorphism of C677T was detected by using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).Hamilton rating scale for depression-24 items(HAMD-24) was used to evaluate depressive symptoms.Results There were significant differences in the distribution of genotypic frequency and allele frequency between the study group and the control group(P0.05).The plasma Hcy levels in subjects with TT genotype in study group and control group were significantly higher than those with CT or CC genotype.Score of HAMD in patients with TT genotype in study group was significantly higher than those with other two genotypes(P0.05).Logistic regression analysis found that the Hcy genotype interaction was positively related with depression.Conclusion TT genotype of MTHFR gene may be the risk factor of the susceptibility of depression.It may affect the severity of depression by influenting the Hcy level.
