Effect of Cordyceps Sinensis on Expression of p-Smad2/3 in Renal Interstitial of UUO Rats

Abstract

Objective To observe the expression of p-Smad2/3 and TGF-β1 in renal interstitial of UUO rats after treatment with Cordyceps sinensis(Cs-4).Methods Fiftyfour male Sprague-Dawley rats were randomly divided into three groups: sham-operated group,UUO model group and Cs-4-treated group.Six rats of each group were sacrificed at the 7th,14th,and 21th days after operation.Histological changes were observed by HE,PAS and Masson staining.The expressions of p-Smad2/3,COL-IV and TGF-β1 protein were detected by immunohistochemistry,and the expression of TGF-β1 mRNA was observed by RT-PCR.Result The index of renal interstitial fibrosis,the expressions of p-Smad2/3,COL-IV and TGF-β1 protein in the renal interstitial,and the expression of TGF-β1 mRNA in renal tissue were decreased significantly in the Cs-4-treated group than those in the UUO model group.Conclusion In UUO rats,Cs-4 may postpone renal interstitial fibrosis by inhibiting the TGFβ-depended Smad signal pathway.
