Establishment of Paper Electrophoresis Determination of γ-aminobutyric Acid and Glutamic Acid in Tea Leaves

Abstract

Due to the high costs in traditional determination of γ-aminobutyric acid(GABA) and glutamic acid(Glu),development of a rapid and economic way for tea leaf production enterprises is important.Paper electrophoresis determination of GBA and Glu was developed and the efficiency was compared to that obtained by means of amino acids analyzer at the concentrations range of 5-30 nmol/μl.The concentrations of GABA which ranged from 5 nmol/μl to 30 nmol/μl as well as Glu correlated linearly with the OD values at 520 nm wavelength with the correlation coefficients of 0.999 5 and 0.999 8,respectively.The average recovery rates of GABA and Glu determined by using paper electrophoresis were up to 101.293% and 101.445%,suggesting this new method worked credibly.The variation coefficients of the contents of GABA and Glu determined by the method were less than 5%,indicating the precision of paper electrophoresis.There was no significant difference in the contents of GABA and Glu determined by paper electrophoresis and amino acid analyzer.
