Abstract
Objective:To study the effects of Keaixin on the proliferation,apoptosis and level of cytokine in tumor-bearing mice.Methods:The cytotoxicity of Keaixin on human hepatoma carcinoma cell line SMMC-7221,human breast carcinoma MDA231 and lung adenocarcinoma A549 cell lines were determined by cell counting kit-8(CCK-8)assay.The morphological changes of the apoptosis cells were observed by transmission electron microscope.The inhibitory effect of Keaixin on tumor growth was observed by the models of implanted sarcoma 180(S180)and hepatoma 22(H22)in mice;The levels of IL-2 and IL-12 in tumor-bearing mice was investigated by Elisa assay.Results:Keaixin have no effect on the proliferation of SMMC-7721、MDA231 and A549 cells.SMMC-7221 cells apoposis was morphologically observed after which were induced by Keaixin.At dosage of 150 and 300mg·Kg-1,the inhibitory rate of Keaixin is 38.52% and 47.88% for S180,32.17% and 40.18% for H22,respectively.Keaixin can increase spleen index and thymus gland index,and enhance the expression levels of IL-2and IL-12.Conclusions:Keaixin have no effect on the proliferation of cells.Keaixin can induce their apoptosis.Keaixin exhibited significantly anti-tumor effect in vivo and the mechanism may be related to enhancing the level of cytokine.
